Title: Experimental study of anti-tumor effects of polysaccharides from Angelica sinensis Key findings: total polysaccharide isolated from Angelica sinensis (Oliv.) Diels (Danggui) was found to increase the survival time of mice bearing leukemia (L1210); decrease the thymus weights of the mice bearing sarcoma (S180); reduce the production of ascitic liquids and prolong the life of mice bearing Ehrlich ascitic cancer. The experiments found: ethanol or aqueous extracts of the herb dose-dependently inhibited the human promyelocytic leukemia cancer cell growth in MTT assay. Flow cytometry analysis revealed that it elicited significant production of sub-G(1) population of the cells, indicating the extract-evoked cell apoptotic death. CONCLUSION: Our results show that the ethanol extract of the herb effectively evokes cancer cell apoptosis, possibly through burst-mediated caspase activation. Key findings: The experiments show: wogonin, derived from the popular Chinese herb Huang-Qin, attenuates NF-kappaB activity by shifting TNFalpha-induced free radical .O(2)(-) to a more reduced nonradical product, H(2)O(2), and thereby sensitizes TNFalpha-resistant leukemia cells to TNFalpha-induced apoptosis.
